Navega Therapeutics is a preclinical-stage biotechnology company that develops epigenetic gene therapies to treat chronic pain and other complex diseases [2,4,6]. The company utilizes a proprietary, AI-enabled zinc-finger epigenome regulation platform to modulate gene expression at the DNA level without making permanent genomic edits [2,3,5,6]. Its lead program, NT-Z001, specifically targets the NaV1.7 sodium channel, a key mediator of pain sensing, to provide a long-lasting, non-addictive alternative to traditional analgesic treatments [2,3,7]. By transiently repressing NaV1.7 expression, Navega aims to replicate the pain-insensitivity phenotype observed in individuals with specific genetic loss-of-function mutations, offering a highly specific therapeutic approach for conditions such as primary erythromelalgia, small fiber neuropathy, and chemotherapy-induced peripheral neuropathy [2,3,4,8].
These therapies are designed to address significant unmet medical needs, specifically providing a non-addictive solution to reduce reliance on opioids and improve the quality of life for patients suffering from chronic pain [3,4,5,8]. The company, founded in 2018 and based in San Diego, is supported by various grants and collaborations, including funding from the National Institutes of Health (NIH) through the HEAL Initiative and the National Cancer Institute, as well as a research collaboration with the National Center for Advancing Translational Sciences (NCATS) [1,3,4,5,7,8]. Ongoing development efforts focus on advancing these therapies into the clinic, including efforts to reformulate NT-Z001 for more accessible administration routes such as localized joint injections [2,4,7].
