Drosophila platform for screening sleep and circadian bioactives

Validated Drosophila-based screening service for evaluating dietary bioactives and botanical extracts on sleep and circadian biology. The platform delivers quantitative behavioral endpoints, scalable throughput, and mechanistic depth to connect bioactive interventions with functional outcomes in disease-relevant models.

Overview

This offering provides a validated Drosophila screening platform for evaluating sponsor-provided dietary bioactives and complex botanical extracts for their effects on sleep and circadian biology. The platform delivers quantitative behavioral endpoints including sleep amount, sleep onset, sleep consolidation, waking activity, and circadian rhythmicity, enabling rapid, scalable assessment of bioactives in a non-vertebrate in vivo system with conserved sleep and circadian mechanisms.

A key advantage is the ability to deliver hydrophobic bioactives as emulsions, taking advantage of the fly's surface-feeding behavior to reduce solvent-related confounds and accelerate dose testing. Initial screens identify behavioral and circadian effects, after which selected candidates can advance to video-based analysis with arousal-threshold testing, as well as functional assays of plasticity, memory, motivated behavior, sleep depth, and brain clearance. Genetic and neuronal tools provide additional mechanistic resolution for promising candidates.

Technical specifications

Core screening capabilities:

  • Quantitative measurement of sleep amount, onset, consolidation, waking activity, and circadian rhythmicity
  • Emulsion-based delivery of hydrophobic bioactives to reduce solvent-related confounds
  • Video-based arousal-threshold testing for sleep depth assessment
  • Functional assays including plasticity, memory, motivated behavior, and brain clearance
  • Genetic and neuronal tools for mechanistic resolution
  • Survival and overt behavioral suppression monitoring

Throughput and scale:

  • 98-monitor Drosophila system with capacity for approximately 3,136 flies per weekly run
  • Allocation of half capacity enables evaluation of approximately 98 compound-dose conditions per week using 16 flies per condition
  • Actual throughput depends on number of doses, controls, administration schedules, and replication requirements

Validation approach:

  • Staged screening and validation program using sponsor-provided bioactives or complex extracts
  • Initial screens quantify behavioral and circadian endpoints
  • Active candidates replicated across dose and time of administration
  • Selected candidates advance to advanced functional and mechanistic studies
Technology readiness level

The platform is fully operational and has been validated through prior demonstration that Gaboxadol acts as a bona fide sleep-enhancing intervention rather than a nonspecific suppressor of activity. Notably, Gaboxadol-induced sleep restored memory in flies expressing human Alzheimer's disease-associated proteins, establishing that the platform can connect bioactive-induced changes in sleep with meaningful functional outcomes in disease-relevant models. The laboratory has prior experience collaborating with industry using externally supplied compounds, and the screening infrastructure is ready to accommodate sponsored research, pilot engagements, fee-for-service contracts, and co-development partnerships.
