A research team from the University of Alberta led by Dr. Frank Wuest has synthesized a new compound for use in peptide cyclization and as a bifunctional chelating agent in the radiolabelling of peptides. Preliminary results using a model peptide indicate that said compound is effective in peptide cyclization by selectively reacting with cysteine residues. A metal-binding DOTAGA moiety in the compound mediates its chelating properties. To facilitate radiolabeling, the compound readily forms stable complexes with metals, including Cu, Dy, Er, Ga, Gd, In, La, Lu, Sc, and Y. As proof-of-concept, the functionality of our compound has been demonstrated using octreotide as the peptide of choice, but may be applied to all cyclic peptides containing a disulphide bridge.
