Introducing a novel bifunctional drug, RS-MNF, that targets GRP55 inhibition and beta2-adrenergic receptor activation to reduce glycolysis in pancreatic cancer, improving therapy efficacy. Validated in multiple models, it utilizes AI for biomarker-guided treatment.

Overview

The innovative bifunctional drug, RS-4'-methoxynaphthylfenoterol (RS-MNF), offers a promising approach to treat pancreatic cancer by targeting two critical pathways: GRP55 inhibition and beta2-adrenergic receptor activation. This dual action reduces glycolysis in pancreatic cancer tumors, thereby enhancing the efficacy of chemotherapeutic and immune therapies. By increasing the pH of the tumor microenvironment (TME), RS-MNF overcomes intrinsic and acquired resistance, providing a robust therapeutic advantage.

Technical specifications

RS-MNF belongs to a family of 18 naphthylfenoterols, specifically designed to selectively inhibit GPR55 and activate the beta2-adrenergic receptor. This compound has demonstrated activity across seven pancreatic ductal adenocarcinoma (PDAC) cell lines and is effective in three mouse models, including PANC-1-derived xenograft, orthotopic PDX, and PDK-derived xenograft. Supported by an NIH grant, these studies have identified serum biomarkers, such as metabolites, proteins, and miRNAs, which can be utilized for treatment assessment and guidance. Future plans include the optimization of dosing protocols and the development of AI-assisted biomarker pattern recognition to enhance treatment precision.

Technology readiness level

The technology is currently at TRL 4, with established efficacy in PDAC models and initial pharmacokinetic and toxicity studies completed. Future validation efforts will focus on optimizing dosing protocols and finalizing pre-IND toxicity studies, projected to be completed within one year.

About PAZ Pharma

PAZ Pharma is a biotechnology company founded in 2019 and headquartered in Mullica Hill, New Jersey, that focuses on the development of novel therapeutic candidates for the treatment of colon cancer. The company specializes in identifying and exploiting the unique metabolic vulnerabilities of cancer cells, aiming to disrupt pathways that are critical for tumor growth and survival. By targeting these specific metabolic mechanisms, the company intends to develop more effective and potentially less toxic treatment options compared to traditional therapies. The research and development pipeline includes both preclinical and clinical-stage candidates designed to address these distinct biological targets.

This work is significant for patients and healthcare providers looking for targeted interventions that move beyond conventional oncology approaches. By concentrating on tumor metabolism, PAZ Pharma seeks to solve the problem of drug resistance and toxic side effects often associated with standard cancer treatments. The organization operates out of the South Jersey Technology Park and focuses its efforts on advancing its lead compounds through critical stages of pharmaceutical development. Through its focused research model, the company aims to contribute to the evolving field of oncology by providing specialized, metabolically-targeted care.
