This innovative solution leverages a CRISPR-Cas12a-based system for the sensitive detection of Salmonella. Utilizing DNA-functionalized gold nanoparticles (AuNPs) and fluorescein isothiocyanate (FITC), it offers a rapid, amplification-free method ideal for complex matrices like cocoa powder. The system addresses detection challenges posed by low pathogen concentrations, making it highly valuable for the food safety industry.
Technical specifications
- CRISPR-Cas12a: Utilized for its precision in targeting Salmonella-specific genes, enabling the cleavage of single-stranded DNA (ssDNA) that links AuNPs and FITC.
- Gold nanoparticles (AuNPs): Synthesized in sizes 20 nm (AuNP20) and 60 nm (AuNP60), they are functionalized with ssDNA and enhance FITC fluorescence through metal-enhanced fluorescence (MEF).
- DNA isolation: Silica magnetic beads (MNPs) are employed for efficient DNA isolation.
- Fluorescence detection: In the presence of target DNA, CRISPR-Cas12a activation leads to ssDNA cleavage, increasing the distance between FITC and AuNP60, thereby restoring fluorescence and enhancing detection sensitivity.
