Mechanistic readouts are supported by EpiBiotech's proprietary Hair.I./EPIGene single-cell transcriptomics platform, enabling pathway-level interrogation (Wnt/β-catenin, TGF-β/fibrosis, JAK-STAT, hypoxia signaling) to link efficacy signals to underlying biology. Key metrics include hair follicle elongation rate and anagen prolongation (ex vivo HFOC), DPC viability and proliferation, spheroid size and trichogenic marker expression, in vivo hair regrowth scoring and quantitative coverage/density imaging, and follicle counts via histology (anagen:telogen ratio). Validation plans incorporate positive controls (e.g., minoxidil, finasteride where applicable), dose-response design, adequate biological replicates, blinded scoring, and reproducibility across independent batches. Molecular validation cross-checks phenotypic endpoints against transcriptomic signatures and qPCR/immunostaining of key markers, ensuring efficacy claims are mechanistically substantiated and translation-ready for partner development programs.
