Intestinal epithelial cotransporter activity assessment and regulation technology

Technology
In development
University

This technology offers a method for assessing and regulating intestinal epithelial cotransporter activity using selective blockers, enabling insights into epithelial function and solute absorption. It utilizes electrophysiological measurements under controlled conditions.

Overview

The intestinal epithelial cotransporter activity assessment and regulation technology provides a novel method for analyzing the acute regulation of sugar and amino acid cotransporters in intestinal mucosae. Using electrophysiological techniques, this technology evaluates epithelial ion transport and resistance, offering insights into altered cotransporter activity and epithelial function. This solution is critical for understanding solute absorption mechanisms and their regulation under various conditions.

Technical specifications
  • Utilizes Ussing chambers for real-time measurement of epithelial short-circuit current (Isc) and transepithelial resistance (TER).
  • Capable of assessing the effects of selective blockers on the apical or basolateral surfaces of intestinal mucosae.
  • Employs well-characterized transport signatures from rodent intestines and human adenocarcinoma cell monolayers.
  • Allows for the use of various electrolyte buffers to interrogate epithelial transport mechanisms.
  • Proven expertise in using SGLT inhibitors like phloridzin and canaglifozin for inhibiting electrogenic Na+-linked transport.
Technology readiness level

This technology is at TRL 4, indicating that it has been validated in a laboratory environment, with a focus on understanding and regulating epithelial cotransporters through precise electrophysiological measurements.
