Innovative monoclonal antibodies targeting specific cow's milk allergen IgE epitopes for sensitive diagnostics and therapy. Offers high specificity and affinity, stable storage, and scalable production.
Nanchang University presents a breakthrough in allergen-specific diagnostics and immunotherapy using monoclonal antibodies developed from cow's milk allergen-specific IgE epitopes. This platform targets key epitopes in cow's milk allergens, providing high specificity and affinity antibodies for sensitive diagnostics and residue assays. These antibodies are stable for up to six months at 4 °C and hold potential for epitope-specific immunotherapy applications.
The research involved screening cow's milk allergy (CMA) sera and identifying 21 linear epitopes, leading to 10 core peptides. Immunization of mice and subsequent antibody screening resulted in antibodies with a dissociation constant (KD) of 1.2–8.7 nM, demonstrating >95% specificity. These antibodies are capable of pinpointing allergenic peptides altered by ultra-processing, contributing to both diagnostics and mechanistic immunology.
Currently at TRL 4, this technology has been validated at the laboratory scale with plans for further optimization and scale-up. Future validation includes enhancing antibody production consistency, large-scale preparation, purification, and comprehensive quality control testing, ensuring readiness for commercial applications.
