A novel multi-layer tablet system combining sustained-release drug seeds with lipid, effervescent, and swellable membrane layers to solve moisture sensitivity and enable rapid, balloon-like drug delivery upon contact with aqueous media.

Overview

This solution addresses a persistent challenge in pharmaceutical formulation: moisture sensitivity in effervescent tablets. The proposed system is a multi-layer tablet architecture designed to protect active ingredients from humidity during storage while enabling rapid, effective drug release when administered. By integrating sustained-release drug seeds with a lipid coating, an inner effervescent layer, and an outer swellable membrane, the tablets remain stable in dry conditions but transform into low-density, balloon-like swollen pills upon contact with body-temperature buffer solutions. The carbon dioxide generated by the effervescent reaction drives this transformation, providing a novel strategy for fast drug delivery to target tissues.

Technical specifications

Key design features:

  • Sustained-release drug seeds form the core of the tablet
  • A lipid outer layer provides the primary moisture barrier
  • An inner effervescent sublayer contains sodium bicarbonate
  • An outer effervescent sublayer contains ascorbic acid
  • A swellable membrane outer coating composed primarily of poly(vinyl acetate) and purified lignin
  • Upon immersion in buffer solution at 37 degrees Celsius, the tablet sinks briefly then transforms into a swollen pill with density below 1 g/ml
  • Carbon dioxide generation from the effervescent reaction drives rapid structural transformation
  • The system is designed to improve solubility and delivery of poorly water-soluble drugs
Technology readiness level

The project is at an early research stage. Initial formulation studies on effervescent tablet techniques have begun, and the research team has prior published experience in developing drug-encapsulated nanomaterials that supports the proposed approach. Future validation will focus on implementing and testing the described multi-layer formulation method, including evaluating the moisture resistance properties and the balloon-like swelling behavior in buffer solutions at physiological temperature.

About Mahatma Gandhi University

Mahatma Gandhi University is a comprehensive public state university on the 110‑acre Priyadarsini Hills campus in Kottayam, serving a broad network of affiliated colleges across central Kerala. Industry engagement is anchored by the Business Innovation and Incubation Centre, established in 2016 with seed support from the Government of Kerala, which provides pre‑incubation, startup mentoring, and IP services. The MGU Innovation Foundation runs technology‑business incubation facilities and programs that convene bootcamps, broker MoUs, and connect companies with faculty and student entrepreneurs; an MoU with Kerala’s Bio360 Life Sciences Park extends access to the regional cluster and commercialization pathways. Research is sustained by competitive national and state funding from agencies such as DST, DBT, CSIR, and ICMR. A dedicated IP and licensing pathway through BIIC supports patenting, licensing, and startup formation.
