Innovative PEG-PAA hydrogel coatings offer pH-responsive encapsulation and controlled release of probiotics, enhancing cell survival in acidic environments and ensuring precise release at neutral pH.

Overview

The development of pH-regulating PEG-PAA hydrogel coatings presents a groundbreaking approach to enhancing the survivability and controlled release of probiotic bacteria. These hydrogels are engineered to encapsulate probiotics, offering a pH-buffering environment that ensures cell survival in harsh acidic conditions while allowing for the controlled release of viable cells at neutral pH. This technology promises significant advancements in the delivery and efficacy of probiotic formulations.

Technical specifications

The PEG-PAA hydrogel system leverages the pH-responsive nature of poly(acrylic acid) (PAA), a polyelectrolyte with carboxylic acid groups that can buffer against low pH. At low pH levels, the COOH groups protonate, providing stability to the encapsulated probiotics. At neutral pH, deprotonation occurs, causing hydrogel chains to repel, swell, and release the probiotics. Using a novel surface-initiated polymerization technique, these hydrogels can encapsulate probiotics individually, maintaining nanoparticle sizes under 200 nm to prevent aggregation.

Key features:

  • pH-responsive encapsulation and release
  • Single-cell encapsulation to prevent aggregation
  • Enhanced probiotic survivability in acidic conditions
Technology readiness level

This technology is currently at TRL 3, having been validated in a laboratory environment. Ongoing research focuses on optimizing hydrogel formulations and validating probiotic encapsulation and release in controlled settings. Future validation plans include characterizing the long-term stability and viability of encapsulated cells, as well as the precise release mechanisms at neutral pH.
