University

VariantForge is a modular platform that accelerates herbicide-tolerance trait discovery by integrating computational mutation prioritization, automated DNA assembly, cell-free expression, and plant-relevant protoplast screening. It reduces time and manual effort versus conventional cloning-expression pipelines, with modules available at TRL 3-9.

Overview

VariantForge addresses the critical bottleneck in engineering herbicide-tolerant traits: the gap between computationally designed protein variants and physically expressed, quality-checked proteins ready for screening. This modular platform combines computational mutation prioritization, automated DNA assembly, cell-free expression, and protoplast-based plant-relevant screening into a scalable workflow that moves from sequences to testable recombinant proteins at high throughput. It directly supports parallel processing in 96/384-well formats and measurably reduces time and manual effort relative to conventional cloning-expression pipelines, enabling rapid iteration of design–build–test cycles.

Technical specifications

Core capabilities:

  • Computational mutation prioritization to generate variant libraries
  • Automated DNA assembly with quality control for rapid construction of selected variants
  • Miniaturized cell-free expression for scalable, parallel protein production (triaging candidates without slow microbial culture cycles)
  • Multiwell protoplast assays to evaluate herbicide tolerance and protein activity under plant-relevant conditions
  • Closed-loop data integration that uses functional results to refine future mutation prioritization, increasing hit rates over successive rounds

Modules can operate independently or as a connected end-to-end workflow, allowing incremental adoption. Performance metrics include throughput, turnaround time, reproducibility, protein quality, and manual effort compared to conventional workflows.

Technology readiness level

The platform spans TRL 3-9. Core modules (computational design, DNA assembly, cell-free expression, protoplast confirmation) are at TRL 5-9, while newer automation is at TRL 3-4. Validation is planned in phased modules, with benchmarking against conventional cloning and expression workflows and progressive integration into an automated end-to-end platform for scale-up.
