Innovative nanoparticle delivery system targeting breast cancer using DOTAP-based LNPs to deliver p53 mRNA and EGFR siRNA. Features transferrin ligand for tumor targeting and pH-sensitive nanoparticles for effective payload release in cytosol.

Overview

This technology introduces a novel delivery system designed specifically for targeting breast cancer cells with p53 mRNA and EGFR siRNA. The system employs DOTAP-based lipid nanoparticles (LNPs) to encapsulate the therapeutic nucleic acids, enhanced with a transferrin ligand to ensure targeted delivery to tumor cells overexpressing transferrin receptors. The pH-sensitive nanoparticles dissolve in the endosomes, facilitating the release of mRNA and siRNA into the cytosol, thereby bypassing the nucleus for a rapid therapeutic effect.

Technical specifications
  • Composition: DOTAP-based LNPs with cholesterol, surface-anchored transferrin, biotin-PEG, and alpha-ketoglutarate.
  • Mechanism: Utilizes electrostatic interactions to protect nucleic acids from nuclease degradation. The hydrophilic outer shell prevents non-specific interactions, enhancing stability and targeting efficiency.
  • Targeting: Transferrin acts as a ligand for active targeting of breast cancer cells.
  • Payload Release: pH-sensitive nanoparticles induce osmotic pressure, causing endosomal rupture and cytosolic delivery of therapeutic agents.
Technology readiness level

The technology is at TRL 4, indicating that it has been validated in a laboratory setting. Future plans include further validation of the hybrid nanocomplexes in vitro and in vivo, assessment of biodistribution, targetability, and therapeutic efficacy in mouse models, as well as biocompatibility evaluation across various dosages.
