RevivBio integrates computational protein design with massively parallel droplet microfluidics to engineer enzymes with enhanced activity, stability, and expression. The platform screens approximately 100,000 variants per campaign, including designed libraries, environmental microbes, and metagenomes, directly at target conditions such as elevated temperature or with plant enzymes. Applications include herbicide-tolerance traits, bioremediation, and industrial enzyme development.

Overview

RevivBio's integrated platform combines computational protein design with high-throughput cell-free functional screening to accelerate enzyme discovery and optimization. It addresses the multi-parametric nature of enzyme engineering—catalytic activity, thermostability, half-life, and expression level—by selecting variants directly on function. Screening approximately 100,000 candidates per campaign, the platform cuts multi-month engineering cycles down to weeks.

Applications include herbicide-tolerance traits, enzymatic bioremediation, and industrial enzyme development. The platform may also be deployed at customer sites with forward-deployed engineers to operate and transfer the technology.

Technical specifications

The platform integrates two complementary capabilities:

  • A computational pipeline using structure-based modeling, homology modeling, inverse folding, and molecular dynamics to design and prioritize enzyme variants
  • Massively parallel droplet microfluidics for functional screening of up to 10^5 candidates per campaign

Additional technical features:

  • Direct screening of environmental microbes, metagenomic DNA libraries, and random or computationally designed variant libraries
  • Robotics-free assembly of 10^4–10^6-member libraries with cell-free expression on release beads
  • Droplet screening in 3–4 days for activity, thermostability, temperature tolerance, and plant-enzyme constraints
  • Protoplast expression for screening under plant-relevant conditions
  • Next-generation sequencing links sequence to function for rapid computational optimization
  • Multi-parametric data generation co-optimizes developability traits in a single campaign
Technology readiness level

The platform has delivered 50x activity gains in rare protein families, demonstrating its throughput and selection efficacy. Nearly all deployed herbicide-tolerance genes originate from environmental microbes, the exact source this platform is designed to mine.

Modular proof-of-concept projects, each running approximately 4–6 weeks, validate specific capabilities against customer-defined success metrics including activity, tolerance, thermostability, expression, and specificity under field-relevant conditions. POCs run independently or in parallel and cover de novo design, designed-library screening, environmental-microbe screening, metagenome mining, assay development for complex enzyme projects, and plant protoplast screening.

About RevivBio

RevivBio is a biotechnology company that creates next-generation biologics and enzymes by integrating a high-throughput microfluidic experimental platform with advanced AI and machine learning models. The company uses generative AI, quantum mechanics, and molecular dynamics to explore sequence space for protein design, allowing for the hyper-speed development of functional biologics and performant enzymes across markets including human health, crop protection, and animal health. Its technology platform enables the rapid screening of large gene libraries to identify variants with enhanced catalytic activity, stability, and substrate specificity.

The company is currently focused on developing enzymatic bioremediation solutions to address environmental contamination, specifically targeting the degradation of PFAS. Backed by the National Science Foundation, this work aims to provide a scalable, cost-effective, and environmentally sustainable alternative to current energy-intensive remediation methods. RevivBio has participated in programs such as Intel Ignite and NYU’s Endless Frontier Labs, and it actively seeks strategic partnerships to apply its bio-inspired design capabilities to diverse industrial and environmental challenges.
