Revolutionary microneedle technology targets tumors to modulate the tumor microenvironment, enhancing the efficacy of CAR-T and T cell engagers. This approach promotes immunogenic cell death, increases therapeutic T cell infiltration, and overcomes tumor heterogeneity.
This innovative technology leverages dissolving microneedles (dMN) for the localized delivery of a hydrophobic natural immune modulator directly to tumor sites and tumor-draining lymph nodes (TDLN). The microneedles transform the immunosuppressive tumor microenvironment (TME) into an immunostimulatory one, enhancing the efficacy of T cell-based therapies such as T cell engagers (TCE) and CAR-T cells. By inducing immunogenic cell death and activating antigen-presenting cells, this method promotes increased infiltration and functionality of therapeutic T cells within tumors. The result is significant inhibition of tumor growth and metastasis across various solid tumors, improving the outcomes of CAR-T and TCE therapies.
The technology is currently at TRL 4, with ongoing validation through in vitro and in vivo studies. HER2+ breast cancer models are used to assess TME modulation, CAR-T cell infiltration, and immune pathway activation. Future validation includes transcriptomic analysis to confirm changes in immune-related genes and pathways.
