MesoRNA’s MSR-seq™ Laboratory Service provides an end-to-end solution for profiling the small non-coding RNA (sncRNA) system with unprecedented resolution. Conventional small RNA-seq often fails to characterize 90% of nucleic acid activity due to rigid secondary structures, size-selection biases, and complex chemical modifications.

Despite the critical role these molecules play in nearly every biological process, a vast portion of the central dogma remains "invisible" to standard technology. At the core of our methodology is a breakthrough capture hairpin oligonucleotide (CHO) that barcodes and immobilizes RNA molecules on a solid support at the earliest stage of library preparation. This "one-pot" on-bead process eliminates the need for sample precipitation, ensuring the comprehensive capture of the entire sncRNA system—including RNA expression, fragmentation, aminoacylation (charging), and modifications—in a single library. The MSR-seq™ platform has been extensively validated to ensure superior precision and breadth (Watkins et al. 2022). Technically, the platform maintains high fidelity across a wide dynamic range, with a robust quantitative correlation (r^2 = 0.94) for tRNA isodecoder expression from inputs as low as 10 ng. Accuracy was further confirmed by measuring stoichiometric ligation efficiency across all four 3’ end nucleotides and utilizing poisoned primer extension to verify (m3C) modification signatures.
