A molecular method utilizing Near Full Genome qPCR to confirm the presence of full-length tobamovirus RNA, enhancing diagnostic accuracy for viral infections. Offers high sensitivity and specificity for detecting intact viral RNA.
The Near Full Genome qPCR is an innovative molecular diagnostic method designed to confirm the presence of full-length tobamovirus RNA. This approach is crucial for assessing the risk of infection, as even low levels of intact RNA can be infectious. The method combines the principles of Near Full Genome PCR with TaqMan qPCR assays, allowing for precise confirmation of full-length RNA. By using specific primers and qPCR amplicons, the presence of fragmented or intact RNA can be determined, providing a reliable indication of infection potential.
Currently, the technology is at TRL 3, with initial trials conducted on leaf material. Future validation plans include optimizing RNA isolation for seeds, enhancing controls and cDNA synthesis, and assessing diagnostic sensitivity and performance. These steps aim to validate the method's specificity, sensitivity, selectivity, repeatability, and reproducibility.
Naktuinbouw, formally known as Stichting Nederlandse Algemene Kwaliteitsdienst Tuinbouw, is an independent quality service organization for the horticulture sector based in Roelofarendsveen, Netherlands. It operates as an autonomous administrative authority (Zelfstandig Bestuursorgaan) under the supervision of the Ministry of Agriculture, Fisheries, Food Security and Nature. The organization is structured into three primary departments—Inspections, Laboratories, and Variety Research—which collectively work to monitor, promote, and guarantee the quality, identity, and health of propagation material such as seeds, cuttings, and young plants. By conducting laboratory testing for diseases and pests, performing DUS (Distinctness, Uniformity, and Stability) research for variety registration, and executing mandatory and voluntary inspections, Naktuinbouw ensures that horticultural products meet rigorous national and international standards.
These services are critical for horticultural entrepreneurs, as they allow producers to reliably trade high-quality plant materials globally while adhering to legal requirements and phytosanitary regulations. In addition to its regulatory and inspection roles, Naktuinbouw functions as a knowledge center that provides training and education to professionals in the green sector. By working in close cooperation with governments, entrepreneurs, and international organizations, the foundation fosters innovation and contributes to a sustainable global food supply. Its role as an accredited, independent body reinforces the reputation of the Dutch horticulture industry for delivering reliable and healthy planting materials, ultimately solving the need for quality assurance in international trade and agricultural compliance.
