Company

PathCRISP offers a field-ready diagnostic platform using CRISPR-Cas12 and PMA/EMA treatment to detect viable Tobamoviruses in seeds. It differentiates between viable and non-viable viruses, providing accurate seed health certification and disease surveillance.

Overview

PathCRISP is an innovative diagnostic platform designed to detect viable Tobamoviruses in seeds using a CRISPR-Cas12-based system. It leverages Propidium Monoazide (PMA) or Ethidium Monoazide (EMA) treatment to selectively block amplification of RNA from non-viable viruses. This method ensures only infectious viruses are detected, offering a significant improvement over traditional qPCR techniques. The system is field-adaptable and provides easy-to-read results via fluorescence or lateral flow immunostrips, making it ideal for seed health certification and disease surveillance.

Technical specifications

Key features:

  • Utilizes CRISPR-Cas12a for precise detection of Tobamoviruses.
  • PMA/EMA treatment differentiates between viable and non-viable viral RNA.
  • Isothermal amplification enhances the speed and efficiency of detection.
  • Modular design suitable for laboratory and field applications.
  • Visual detection through fluorescence or lateral flow immunostrips for ease of use.
Technology readiness level

This diagnostic platform is currently at Technology Readiness Level 6, having been validated in relevant environments with further testing planned. The development process includes five key phases over eight months, culminating in a portable kit with lyophilized reagents to enhance shelf life and usability.
