What It Is: A proprietary, patent-protected dual-pathway metabolic trait platform enabling food crops to autonomously biosynthesize and accumulate both Provitamin D3 (7-dehydrocholesterol) and active Coenzyme B12 (adenosylcobalamin) within edible tissue. Protected under U.S. Patent Application No. 64/148,820.

Why It Matters: Vitamin D3 and Vitamin B12 deficiency affects over 1 billion people worldwide, particularly in plant-based and vegetarian populations where staple crops cannot naturally provide these essential nutrients. Previous academic attempts to engineer Provitamin D3 using total CRISPR knockouts resulted in severe brassinosteroid starvation, causing dwarfism and yield penalties. Our platform overcomes this biological bottleneck while preserving full vegetative vitality.

How It Works:

  1. Provitamin D3 Pathway: Utilizes a kinetically tuned plant reductase designed to attenuate catalytic turnover by approximately 85%. This preserves 15% vital flux for downstream phytosterols and vegetative growth (preventing stunting), while shunting precursor accumulation to deliver 18.5 µg Provitamin D3 per 100g fresh edible serving (100% daily adult RDA).
  2. Active Coenzyme B12 Pathway: Overcomes the plant kingdom barrier by introducing a chloroplast-targeted aerobic cobalt-chelation machinery driven by an endogenous transit peptide, driving autonomous accumulation of 3.5 – 4.5 µg active Coenzyme B12 per 100g fresh edible serving (150% daily adult RDA).

Proof to Date (In-Silico & Computational De-risking Complete):

  • Molecular modeling and structural docking validated via ESMFold/AlphaFold3 (Delta G = -10.24 kcal/mol, pLDDT > 94/100).
  • Supercomputer sequence alignment verified via NCBI BLASTX with >99.3% and >99.6% identity against functional catalytic backbones, confirming 100% in-frame translation.
  • Broad phylogenetic compatibility validated across Solanaceous species (tomato, bell pepper, eggplant, potato), cereal grains, and aquatic biomass (duckweed).
  • Synthesis-ready, plant-optimized cDNA constructs prepared with standardized flanking restriction cassettes for rapid modular cloning.

Next Steps & Collaboration Objectives: Phase 1 computational design and patent locking are complete. We are seeking collaborative industrial partners (ag-biotech leaders, seed developers, and food conglomerates) for Phase 2 in-planta screening:

  • High-throughput transient leaf infiltration (Nicotiana benthamiana) and HPLC-DAD / LC-MS quantification.
  • Stable transformation pipelines in target commercial crop cultivars under co-development, sponsored research, or commercial trait licensing agreements.
