Novel dual-action antifungal agents combining SDH and Qo inhibitory pharmacophores to overcome drug resistance. Built on fluorine-substituted pyrazole amide cores with methoxyacrylate motifs, designed using fragment-based drug design and validated through molecular modeling.
This research program develops novel dual-target antifungal agents that simultaneously inhibit succinate dehydrogenase (SDH) and coenzyme Qo, two critical targets in commercial fungicides. By combining pharmacophores from both SDH inhibitors (SDHIs) and Qo inhibitors (QoIs) into single molecules, the approach addresses the growing problem of fungicide resistance that limits the effectiveness of single-target drugs. The work builds on prior findings that fluorine-substituted pyrazole amide compounds exhibit strong antifungal activity and aims to extend this foundation by integrating methoxyacrylate-type active groups characteristic of QoIs.
The program is at an early-to-mid research stage. Molecular modeling studies have provided initial support for the dual-target design rationale. Future work will establish a candidate compound library, synthesize and screen novel SDH/Qo dual-target inhibitors, and evaluate their antifungal potential and resistance profiles. The goal is to lay the groundwork for the development of next-generation fungicides with reduced resistance risk.
