Ningbo University presents a novel methodology for discovering and screening small molecule-drug conjugates (SMDCs) specifically targeting Pancreatic Ductal Adenocarcinoma (PDAC). By utilizing chemoproteomics probes, the technology aims to selectively recognize and internalize these conjugates into PDAC cells. This approach promises to improve the specificity and efficacy of cancer treatments by ensuring that therapeutic agents are accurately delivered to malignant cells, minimizing off-target effects.
The core of this technology lies in a library consisting of approximately 4000 photoaffinity probes and 300 covalent probes based on drug-like molecular scaffolds. These probes are designed with terminal alkyne groups, allowing for rapid 'click' reactions with fluorescent molecules. This facilitates the visualization and identification of PDAC-specific membrane antigens through fluorescence microscopy. The process involves competitive assays to refine and select high-affinity ligands for PDAC, followed by the design of SMDCs that demonstrate potent internalization selectivity.
This technology is currently at TRL 3, indicating that analytical and experimental critical function and/or characteristic proof of concept has been achieved. The upcoming validation steps are expected to be completed within 4-6 months, further advancing the development and application of this promising cancer treatment technology.
