The barrier has been technical: tRNAs are notoriously difficult to reverse transcribe due to their structure and dense modifications, so standard small RNA-seq systematically misses them. MSR-seq solves this. It captures full-length tRNAs and their fragments in a single workflow while preserving the modification signatures (such as m1A m22G, and PseudoU) that govern tRNA function and processing, opening a high-abundance regulatory layer that has been effectively invisible to conventional methods.
